Concentration: 1000U/ul.
Definition of activity: 1 active unit refers to the amount of enzyme needed to increase the 260nm wavelength absorbance of yeast RNA by 15 minutes at 37 ºC and pH7.5.
Enzyme activity analysis mixture: 50mM Tris-HCL (PH7.5), 2mM EDTA, 3mg/ml yeast RNA.
Preservation buffer composition: 50mM Tris-HCL (PH7.4) and 50% (vmaw) glycerol.
Quality control: related tests showed that there was no contamination of endonuclease and exonuclease. The functional test method was RNA degradation during the purification of plasmid DNA (synergistic effect with RNase A).
Inhibitors: metal ions Mg2+, Ca2+, Zn2+, Fe2+, Cu2+ (inhibition efficiency of about 40% at MgCL2, 100mM concentration; inhibition efficiency of about 30% at 10mm concentration of 1mM), single nucleotides (2lam, GMP, GMP, etc.), guanilyl-2-5-guanosine.
Inactivation: thermal inactivation is reversible. It is recommended to remove the enzyme by column purification / extraction.
Character: ribonuclease T1 is an endonuclease that specifically degrades single-stranded RNA at G residue site.
The enzyme cleaves the phosphodiester bond between the 3'- guanosine residue and the adjacent nucleoside 5'-OH group by forming a nucleoside 2-amino-3-cyclic phosphate intermediate.
The reaction product is 3'-GMP terminal oligonucleotide.
RNase T1 is active without the involvement of metal ions.
