The sample processing is as follows: the sample is mixed with Fermentas 6×DNA Loading Dye&SDS solution (#R1151), heated at 75°C for 5 minutes or 65°C for 10 minutes and stored in an ice bath; during transformation, the volume of the ligation product must not exceed the volume of competent cells 10%; before electrotransformation, remove the T4 DNA Ligase in the ligation mixture by extraction method, and then purify the extraction product by ethanol precipitation
